Multi-omics integration reveals the dynamic impacts of stock enhancement on the intestinal health condition in kaluga sturgeon (Huso dauricus)

(Submitter supplied) On October 6th,2023, one thousand kaluga sturgeon were released to Songhua River (119°52′-132°31′ E,11°12′-51°38′ N) after 2 h adaptive domestication to eliminate the physiological response caused by transport stress. Before wild reintroduction to river, non-toxic fluorescent dye was injected into the snout of kaluga sturgeon, which was beneficial to catch them again. On the 0 d (Ori), 7 d (D7FL), 14 d(D14FL) and 30 d (D30FL), three kaluga sturgeon individuals were recaptured randomly using seine nets. more...

PFKFB3 drives renal fibrosis through promoting histone lactylation-mediated NF- B activation [RNA-seq]

(Submitter supplied) Persistently elevated glycolysis in kidney has been demonstrated to promote chronic kidney disease. However, the underlying mechanism remains largely unclear. Here, we observed that PFKFB3, a key glycolytic enzyme, was remarkably induced in kidney proximal tubular cells following ischemia-reperfusion injury in mice, as well as in multiple etiologies of chronic kidney disease patients. Proximal tubular epithelial-specific deletion of PFKFB3 significantly reduced renal lactate levels, mitigated inflammation and fibrosis, and preserved renal function in the IRI mouse model. more...

Targeting senescence induced by age or chemotherapy with a polyphenol-rich natural extract improves longevity and healthspan in mice

(Submitter supplied) The accumulation of senescent cells within tissues contributes to age-related diseases through the senescence-associated secretory phenotype (SASP). To fight senescence and maintain a healthy tissue microenvironment, the quest for longevity-promoting compounds has resulted in promising discoveries from botanical sources. Here we show that daily administration with a low dose of Haenkenium (HK), a standardized extract of Salvia haenkei, extended the lifespan and healthspan of aged C57BL/6 mice by delaying senescence and consequently ameliorating systemic inflammation and fibrosis markers in multiple tissues including skin, muscle, cartilage, and kidney. more...

Embryonic myeloid cells orchestrate niche cell homeostasis critical for the establishment of the definitive HSC pool [II]

(Submitter supplied) The lifelong replenishment of blood cells relies on the function of definitive HSCs that migrate to the bone marrow during development. This process is tightly controlled by the bone marrow microenvironment. Embryonic macrophages emerge before the onset of definitive hematopoiesis, seed into discrete tissues and contribute to specialized resident macrophages throughout life. However, the functional impact of embryonic macrophages on HSCs or the niche remains unknown. more...

Molecular characterization and differential expression profiles of transcription factor Sox gene family between ovaries and testes from golden pompano (Trachinotus ovatus)

(Submitter supplied) The SRY-related HMG-box (Sox) is a transcription factor related to the testis-determining gene SRY (Sex determination Region of Y chromosome), which plays an important role in the sexual development of Osteichthyes. In order to understand the characteristics of Sox gene family members in Trachinotus ovatus(ToSox), this study used bioinformatics methods to systematically analyze the physicochemical properties, phylogenetic tree, chromosomal localization, and prediction of interacting proteins of the ToSsxs. more...

Endothelial IGFBP6 Suppresses Vascular Inflammation and Atherosclerosis

(Submitter supplied) It has previously been reported that expression levels of insulin-like growth factor binding protein 6 (IGFBP6) are reduced in unstable plaques compared to stable plaques in patients with coronary heart disease (CAD). However, the exact role and mechanism of IGFBP6 in endothelial homeostasis and inhibition of focal atherosclerotic plaque development in vivo remains unclear. Transcripome sequencing and bioinformatics analysis of IGFBP6-overexpressed human umbilical vein endothelial cells (HUVECs) suggest that pathway analysis and differentially expressed mRNA may help clarify the role and potential mechanism of IGFBP6 in the treatment of cardiovascular diseases.

Single-cell RNA-seq analysis of mouse embryonic stem cells before and after Cohesin depletion

(Submitter supplied) The contrast between the disruption of genome topology upon cohesin loss and the lack of downstream gene expression changes instigates intense debates regarding the structure-function relationship between genome and gene regulation. Here, by analyzing transcriptome at the single-cell level, we discover that, instead of dictating population-wide gene expression levels, cohesin supplies a general function to neutralize stochastic co-expression tendency of cis-linked genes in single cells. more...

Aged skin exacerbates experimental osteoarthritis via enhanced IL-36R signaling

(Submitter supplied) To investigate skin aging is an important driver of experimental osteoarthritis(OA) progression in mice via enhanced IL-36 receptor (IL-36R) signaling. The supernatants from the co-culture of UV-aged mouse primary keratinocytes with synovial fibroblasts were collected to induce mouse primary articular chondrocytes (CCs) to become aging SNL CCs. On this basis, Spesolimab (100ng/ml) was added to culture aging SNL CCs to study the drug's effect on OA. more...

RKIP regulates bone homeostasis by mediating the differentiation fate of bone niche macrophages.

(Submitter supplied) As central cells involved in osteoimmunology, bone niche macrophages possess diverse functions, and their differentiation fate regulates bone homeostasis. Elucidation of the underlying mechanism involved in macrophage differentiation is important for developing new therapeutic targets for osteoporosis. Our RNA-seq data showed that the differentially expressed genes in BMMs after RKIP knockout were involved in the pathways of cell adhesion and osteoclast differentiation.
